Lab 5Cellular Respiration Introduction: Cellular respiration is an ATP-producing catabolic process in which the ultimate electron acceptor is an inorganic molecule, such as oxygen. It is the release of energy from organic compounds by metabolic chemical oxidation in the mitochondria within each cell. Carbohydrates, proteins, and fats can all be metabolized as fuel, but cellular respiration is most often described as the oxidation of glucose, as follows: C6H12O6 + 6O2 > 6CO2 + 6H2O + 686 kilocalories of energy/mole of glucose oxidized Cellular respiration involves glycolysis, the Krebs cycle, and the electron transport chain.Glycolysis is a catabolic pathway that occurs in the cytosol and partially oxidizes glucose into twopyruvate (3-C). The Krebs cycle is also a catabolic pathway that occurs in the mitochondrial matrix and completes glucose oxidation by breaking down apyruvate derivative (Acetyl-CoA) into carbon dioxide. These two cycles both produce a small amount of ATP by substra te-level phosphorylation and NADH by transferring electrons from substrate to NAD+ (Krebs cycle also produces FADH2 by transferring electrons to FAD).The electron transport chain is located at the inner membrane of the mitochondrion, accepts energized electrons from reduced coenzymes that are harvested during glycolysis and Krebs cycle, and couples this exergonic slide of electrons to ATP synthesis or oxidative phosphorylation. This process produces 90% of the ATP. Cells respond to changing metabolic needs by controlling reaction rates. Anabolic pathways are switched off when their products are in ample supply. The most common mechanism of control is feedback inhibition.Catabolic pathways, such as glycolysis and the Krebs cycle, are controlled by regulating enzyme activity at strategic points. A key control point of catabolism is the third step of glycolysis, which is catalyzed by an allosteric enzyme, phosphofructokinase. The ratio of ATP to ADP and AMP reflects the energy status o f the cell, and phosphofructokinase is sensitive to changes in this ratio. Citrate and ATP are allosteric inhibitors of phosphofructokinase, so when their concentration rise, the enzyme slows glycolysis.As the rate of glycolysis slows, the Krebs cycle also slows since the supply of Acetyl-CoA is reduced. This synchronizes the rates of glycolysis and the Krebs cycle. ADP and AMP are allosteric activators for phosphofructokinase, so when their concentrations relative to ATP rise, the enzyme speeds up glycolysis, which speeds of the Krebs cycle. Cellular respiration is measure in three manners: the consumption of O2 (how many moles of O2 are consumed in cellular respiration? ), production of CO2 (how many moles of CO2 are produced in cellular respiration? , and the release of energy during cellular respiration. PV = nRT is the formula for the inert gas law, where P is the pressure of the gas, V is the volume of the gas, n is the number of molecules of gas, R is the gas constant, and T is the temperature of the gas in degrees K. This law implies several important things about gases. If temperature and pressure are kept constant then the volume of the gas is directly proportional to the number of molecules of the gas. If the temperature and volume remain constant, then the pressure of the gas changes in direct proportion to the number of molecules of gas.If the number of gas molecules and the temperature remain constant, then the pressure is inversely proportional to the volume. If the temperature changes and the number of gas molecules is kept constant, then either pressure or volume or both will change in direct proportion to the temperature. Hypothesis: The respirometer with only germinating peas will consume the largest amount of oxygen and will convert the largest amount of CO2 into K2CO3 than the respirometers with beads and dry peas and with beads alone.The temperature of the water baths directly effects the rate of oxygen consumption by the contents in the respirometers (the higher the temperature, the higher the rate of consumption). Materials: The following materials are necessary for the lab: 2 thermometers, 2 shallow baths, tap water, ice, paper towels, masking tape, germinating peas, non-germinating (dry) peas, glass beads, 100 mL graduated cylinder, 6 vials, 6 rubber stoppers, absorbent and non- absorbent cotton, KOH, a 5-mL pipette, silicon glue, paper, pencil, a timer, and 6 washers. Procedure:Prepare a room temperature and a 10oC water bath. Time to adjust the temperature of each bath will be necessary. Add ice cubes to one bath until the desired temperature of 10oC is obtained. Fill a 100 mL graduated cylinder with 50 mL of water. Add 25 germinating peas and determine the amount of water that is displaced. Record this volume of the 25 germinating peas, then remove the peas and place them on a paper towel. They will be used for respirometer 1. Next, refill the graduated cylinder with 50 mL of water and add 25 non-germinating peas to it.Add glass beads to the graduated cylinder until the volume is equivalent to that of the expanded germinating peas. Remove the beads and peas and place on a paper towel. They will be used in respirometer 2. Now, refill the graduated cylinder with 50 mL of water. Determine how many glass beads would be required to attain a volume that is equivalent to that of the germinating peas. Remove the beads. They will be used in respirometer 3. Then repeat the procedures used above to prepare a second set of germinating peas, dry peas and beads, and beads to be used in respirometers 4,5,and 6.Assemble the six respirometers by obtaining 6 vials, each with an attached stopper and pipette. Then place a small wad of absorbent cotton in the bottom of each vial and, using the pipette or syringe, saturate the cotton with 15 % KOH. Be sure not to get the KOH on the sides of the respirometer. Then place a small wad of non- absorbent cotton on top of the KOH-soaked absorbent cotton. Repeat the se steps to make the other five respirometers. It is important to use about the same amount of cotton and KOH in each vial. Next, place the first set of germinating peas, dry peas and beads and beads alone in vials 1,2, and 3.Place the second set of germinating peas, dry peas and beads, and glass beads in vials 4,5, and 6. Insert the stoppers in each vial with the proper pipette. Place a washer on each of the pipettes to be used as a weight. Make a sling using masking tape and attach it to each side of the water baths to hold the pipettes out of the water during the equilibration period of 10 minutes. Vials 1,2, and 3 should be in the bath containing water at room temperature. Vials 4, 5, and 6 should be in the bath containing water that is 10oC. After the equilibration period, immerse all six respirometers into the water completely.Water will enter the pipette for a short distance and stop. If the water does not stop, there is a leak. Make sure the pipettes are facing a direction f rom where you can read them. The vials should not be shifted during the experiment and your hands should not be placed in the water during the experiment. Allow the respirometers to equilibrate for three more minutes and then record the initial water reading in each pipette at time 0. Check the temperature in both baths and record the data. Every five minutes for 20 minutes take readings of the waterââ¬â¢s position in each pipette, and record.Results: In this activity, you are investigating both the effects of germination versus non-germination and warm temperature versus cold temperature on respiration rate. Identify the hypothesis being tested on this activity. The rate of cellular respiration is higher in the germinating peas in cold than in the beads or non-germinating peas; the cooler temperature in the cold water baths slows the process of cellular respiration in the both germinating and non-germinating peas. Because of this fact, the peas contained by the respirometers placed in the water at 10oC carried on cellular respiration at a lower rate than the peas in respirometers placed in the room temperature water. Militarization of Our Police Essay
I strongly feel that military tactics, training, and equipment do not have a place in American law enforcement because dozens of needless deaths and injuries have occurred because of it and itââ¬â¢s extremely expensive to supply these things. First, the American law enforcement has become much too aggressive with its citizens. There are several reports of police going above and beyond, using extreme measures, to take down the general public. One perfect example would be the MacDonald case. In the summer of 2006 a frail, troubled 18-year-old girl named Ashley MacDonald ran through a nearly empty Huntington Beach, California, city park in the early morning holding a small knife. An onlooker called the police and soon two large male officers showed up. They shot the girl to death with 18 bullets, claiming she had lunged toward them and put their lives in danger (Greenhut, 2008). Another example showing excessive, military force being used would be the Jose Guerena case. The home of former Marine and Iraq combat veteran Jose Guerena was raided by local sheriffs in Pima County, Arizona, who were attempting to execute a narcotics search warrant. Unfortunately, the manner in which the SWAT team invaded Guerenaââ¬â¢s home prompted Guerena to reach for his gun to protect himself and his family. Sheriffs then shot off 71 rounds and hit Guerena with 22, killing him (Clabough, 2011). Second, it has cost the government millions of dollars to supply the American law enforcement with military equipment, which I believe is completely unnecessary. According to Pentagon data cited by The Daily, the Defense Department gave away almost $500 million worth of surplus military equipment to state and local law enforcement in 2011, which was a new record. And next yearââ¬â¢s orders are already up more than 400 percent (Newman, 2011). The crime rate may have decreased, but the detailed numerous accounts of abuses and failed operations that left innocent people dead and the record billions worth of military equipment still being handed to local police is absolutely ridiculous. I feel there is more bad than good deriving from the militarization of our police.
Les Murrayââ¬â¢s ââ¬ÅThe Widower in the Countryââ¬Â Essay
In ââ¬Å"The Widower in the countryâ⬠, change is conveyed in many ways. The poem shows that change can bring a life of isolation and loneliness. The poem also shows how change is not always embraced and how we should not lead a life with a lack of change. The title of the poem, the widower in the country immediately gives the reader the impression of an individual in a vast area. This lets the readers know that the widower is alone and isolated. Already, the reader already feels sympathy for the widower, not only because he has lost his wife, but he now lives alone in a vast and empty area. In the first stanza, the first line, ââ¬Å"Iââ¬â¢ll get up soon, and leave my bed unmadeâ⬠, shows how the persona leads a life that is monotonous and repetitive each day. There is a certainty and reluctance in his tone and it seems as though he now has no reason to make his bed, as he is lonely and there is no one who will see the bed even if it was made. At the end of the stanza, ââ¬Å"For I get up late nowâ⬠, the word ââ¬Å"nowâ⬠has been deliberately placed to show how the widower has changed his behaviour. In the second stanza, the personification of ââ¬Å"Christmas paddocks, aching in the heatâ⬠, imitates the personas own feelings, and the words ââ¬Å"aching in the heartâ⬠look like aching in the heart, which is what the persona feels like, as he has lost his wife. Christmas is also usually a time for family gathering, and this highlights how change has caused the widower to lead a life of loneliness and isolation. This stanza is also filled with negative imagery and mundane activities- ââ¬Å"The windless trees, the nettles in the yardâ⬠¦ and then Iââ¬â¢ll go in, boil water and make tea.â⬠The â⬠¦ at the end of the sentence shows that the widowers day is filled with other activities that are more or the same, again highlighting his life that has a lack of change. The lack of punctuation in this stanza also reflects the widowerââ¬â¢s monotonous life and again shows how change can bring a life of isolation and loneliness. In the third stanza, there is again a lack of punctuation. ââ¬Å"Iââ¬â¢ll stand out on the hill and watch my house away below, and how the roof reflects theâ⬠.à There is a certainty in the widowerââ¬â¢s tone and the detached images show he doesnââ¬â¢t want to be in this situation. ââ¬Å"Makes my eyes waterâ⬠gives us an image of the widower in misery and crying and we feel sympathy for him. This also shows he has not embraced the change. ââ¬Å"Close on bright webbed visions smeared on the dark of my thoughts to dance and fade awayâ⬠shows how the persona has visions and memories of the past. There is a paradox between bright and smeared and this reflects the personaââ¬â¢s misery. Although he wishes to think of the happier times in the past, they have now become blurred and have faded away. ââ¬Å"Then the sun will move onâ⬠shows the widowers unhappiness and ââ¬Å"I will simply watch, or work, or sleepâ⬠is a cumulative listing of three monotonous and mundane activities, which highlights the lack of variety in the widowerââ¬â¢s life. The sentence also shows how the monotony is never-ending. In ââ¬Å"And evening will come onâ⬠the ââ¬Å"andâ⬠is placed at the start of the sentence to draw attention to how the widower is so certain about his life as it is always the same, day after day. In the last stanza, ââ¬Å"Getting near dark, Iââ¬â¢ll go home, light the lamp and eat my corned-beef supper, sitting there at the head of the tableâ⬠shows how the widowerââ¬â¢s day is filled with even more mundane activities and his reluctance to make any changes. The fact that he is sitting at the head of a table highlights his loneliness, as he has no family, and is by himself. ââ¬Å"Then Iââ¬â¢ll go to bedâ⬠again shows the certainty in his tone and his lack of change. ââ¬Å"Last night I thought I dreamedâ⬠is the only time there is a change in tense and harsh sounding images- ââ¬Å"the screaming was only a possum ski-ing down the iron roofâ⬠follows it. The widower has lost all hope as he cannot even dream and has nothing positive to look forward to, not even in his dreams. The poem ends with its only positive image ââ¬Å"little moonlit clawsâ⬠. With uses of imagery, personification, paradox, assonance and cumulative listing, Les Murrayââ¬â¢s ââ¬Å"The Widower in the Countryâ⬠clearly shows how change can lead to a life of isolation and loneliness.

These economies however have not reached the levels of being able to control the world economy as the United States has managed. There is power in the extent or the area of land and the total population commanded by a state to be considered powerful. The small economies mentioned above have not been able to make it to the global sphere because of their small land sizes as well as their small populations. In terms of military specialization and war dominations, the world has been looking forward to the development of nuclear weapons for purposes of war. Many nations considered to be among the strongest nations in the world have developed very technical and sophisticated war machines. Wang has listed some of these such as South Korea among others. These countries have as well developed very lethal nuclear weapons which have been considered a major source of threat to the entire world (Wang 1999). As a result there have been rumors of wars as well as some other wars fought between various nations of the world. The rumors of wars are what the political analysts and war experts refer to as the cold war and begun some years ago. In order to maintain the economic, political and military power, the united states knows very well that she has to employ certain techniques to enable her survive in her position. Many of the Athenians who supported the democrats and the democrats who were overthrown by the tyrants viewed Criatias actions as emanating from Socrates teachings. The fact that Critias made it clear that he had cast aside the follies of his youth, such as education, did not change the Athenians view of Socrates role in the growth of the tyrants. Soon after, the Tyrants were replaced by the democrats. As a sign of reconciliation and a means of healing the wounds of the civil war Socrates perceived role was sidestepped. However, Anytus who was a leading voice among the democrats wa s not as forgiving. His actions were not only a result of the civil war but were mostly personal as his son was a keen follower of Socrates teachings. ... The first of these charges was impiety. Under impiety, Socrates was accused of believing in multiple gods of which the state never believed in. In extension, he failed to seek divine explanations and instead sought natural explanation for natural phenomenon. The second charge was that he corrupted the young, as part of the charge Socrates was accused of invoking a spirit of criticism among the youth (Fagan and John 121). As part of his regular teachings Socrates received great audience from the youth across all social groups in Athens, he critically evaluated prominent individuals in the marketplace. In his cross-examination of prominent people such as poets, artisans, and statesmen Socrates often dismissed them as lacking in knowledge and believing to know more than they actually did (Brickhouse and Smith 26). It is his ability to criticize those in power that he struck a code with the young and this had a profound effect among them. As stated, Socrates trial was a result of witch h unting and a bulk of what Socrates was accused of was a misrepresentation or misinterpretation of his engagements in Athens. According to the Athenians, piety was a representation of several engagements. Key among these was respect for the gods, the dead and the ancestors. They believed that any impious person was only attracting the wrath of the gods and the entire society would suffer in return, mainly through plague and sterility. Impiety charge was thus a serious charge among the Athenians. Because Socrates was a critique of existent practice it is possible that any of his words or actions would have translated to an impiety charge.
